How to Make Potato Dextrose Agar (PDA) Using Potato Flakes

Potato flakes are an excellent choice for preparing Potato Dextrose Agar (PDA), a widely used medium for agar plates!

Key Properties of PDA Agar Mix:

  • Sterilizes at temperatures above 121Β°C for 90 minutes per 250ml
  • Melts at temperatures over 80Β°C
  • Solidifies at temperatures below -45Β°C

Ingredients for 1000ml of PDA Agar:

  • 2000ml water jug (for mixing)
  • 1000ml boiling water (distilled water is recommended for best results)
  • 20g potato flakes
  • 15g agar powder
  • 10g cane sugar, maple syrup, or regular sugar
  • 5-6 drops of agar colorant (We use blue as it helps highlight contamination more effectively.)

Preparation Method:

  1. Combine all ingredients in the water jug, stirring thoroughly until you achieve a smooth, consistent mixture. The smoother, the better!
  2. Transfer the mixture into sterilization-safe bottles or containers.
  3. Before placing the container in a pressure cooker, ensure the lid is loosely secured to prevent pressure buildup and potential spills.
  4. Sterilize at 121Β°C for 90 minutes to ensure a contaminant-free medium.
  5. Once sterilized, the PDA mix can be stored for future use.
